Harvard Scientists Reverse the Ageing Process

Harvard scientists were surprised that they saw a dramatic reversal, not just a slowing down, of the ageing in mice. Now they believe they might be able to regenerate human organs.
Scientists claim to be a step closer to reversing the ageing process after rejuvenating worn out organs in elderly mice. The experimental treatment developed by researchers at the Dana-Farber Cancer Institute, Harvard Medical School, turned weak and feeble old mice into healthy animals by regenerating their aged bodies….
“What we saw in these animals was not a slowing down or stabilisation of the ageing process. We saw a dramatic reversal – and that was unexpected,” said Ronald DePinho, who led the study, which was published in the journal Nature….
Repeating the trick in humans will be more difficult. Mice make telomerase throughout their lives, but the enzyme is switched off in adult humans, an evolutionary compromise that stops cells growing out of control and turning into cancer. Raising levels of telomerase in people might slow the ageing process, but it makes the risk of cancer soar.
Not exactly true. Do they meaning elites, really want you to know that reversing the ageing process is relatively simple. Telomerase does not cause Cancer cells. Uncontrolled expression of the telomerase gene is a symptom of cancer not a cause. In healthy cells this would not be a problem. Uncontrolled expression of telomerase in healthy cells would just cause the replication of more of the same kind. Cancer is an expression of the malfunction of the replication process not the replication process itself.

The implications of the Geron press release are that telomerase controls the cellular aging clock that triggers either a regeneration into sustained youth or a breakdown into accelerated aging with the loss of telomere lenght. Age reversal may be simply adding back telomerase into the body.

MENLO PARK, Calif.–(BW HealthWire)–Nov. 13, 2000–Geron Corp. (Nasdaq:GERN – news) announced the publication of research demonstrating that the telomerase gene restores the ability of aging human skin cells to form normal skin structures in a mouse model of tissue formation. Published in the journal Experimental Cell Research, the work was conducted by scientists at Geron Corporation and Stanford University.
Age-related changes in skin cells play a role in conditions such as chronic ulcers and photoaging. Skin is composed of two principal cell types: keratinocytes, which form the upper epidermal layer, and fibroblasts, which form the underlying dermal structures. These layers are connected by a tight junctional membrane. The research team discovered that fibroblasts aged in the laboratory lost the ability to form a robust junction with young human keratinocytes when the two cells were put into an animal model of tissue formation. This condition is observed in the elderly and is manifested by increased skin frailty and subepidermal blistering.
In the study, introduction of telomerase to aging fibroblasts dramatically increased their division capacity and restored their ability to reconstitute normal human skin structures in the model system. A genomics microarray analysis also showed that telomerase restored a normal pattern of expressed genes to old fibroblasts. Telomerase, therefore, not only confers replicative immortality to skin fibroblasts, but also prevents or reverses the loss of biological function associated with aging cells.
“This is the first demonstration of a beneficial effect of telomerase activation in human cells in an in vivo animal model,” stated Calvin Harley, Ph.D., Geron’s chief scientific officer. “The research brings the company one step closer to a telomerase gene therapy for the treatment of chronic degenerative diseases in the elderly, including debilitating skin ulcers.”
The two critical genes for human telomerase activity were cloned and characterized by Geron scientists (Science 269, 1236-1241, 1995 and Science 277, 955-959, 1997). Telomerase is an enzyme that maintains telomere length in immortal cells and confers replicative immortality without malignant transformation. Generally, normal human body cells lack telomerase and lose a small amount of telomeric DNA at each cell division, until a threshold length is reached which triggers senescence and loss of function. Critical telomere loss in certain cells at sites of chronic stress in humans contributes in a fundamental way to diseases of the elderly.

Another study has shown that Centarians have an active gene that secrete telomerase more efficiently than non-centarians. Of course they are still subjected to the same aging process and shortening of the telomeres. Telomeres provide information for the DNA to split. As the lenght of Telomeres shortens the rate at which DNA splits slows down and errors crop up more frequently in the code. With saturation of telomerase is should be possible for a complete regeneration of the Genetic code. Researchers are afraid of the implications of their own work. Scientists have zeroed in on one apparent key to long life: an inherited cellular repair mechanism that thwarts aging and perhaps helps prevent disease. Researches say the finding could lead to anti-aging drugs.
The study involves telomeres, the ends of chromosomes that have been likened to the plastic tips that prevent shoelaces from unraveling. Telomeres were already known to play a key role in aging, and their discovery led to this year’s Nobel Prize in medicine.
The new study, which focused on Ashkenazi Jews, finds those who lived the longest had inherited a hyperactive version of an enzyme called telomerase that rebuilds telomeres.
In effect, centenarians tend to have a top-notch body mechanic at work 24/7 repairing the hardware that runs the body, versus a normal person whose body’s cellular control center is left to wear out with time.
“Humans of exceptional longevity are better able to maintain the length of their telomeres,” said Yousin Suh, associate professor of medicine and of genetics at the Albert Einstein College of Medicine at Yeshiva University. “And we found that they owe their longevity, at least in part, to advantageous variants of genes involved in telomere maintenance.”
The results are detailed this week in the Proceedings of the National Academy of Sciences.
Telomeres are short bits of specialized DNA that cap the chromosomes, which tell a cell what to do. Over time, cells divide over and over to keep the body alive. But with each cell division, telomeres get shorter. When they become too short, the cell stops dividing and lapses into a state called cell senescence. Vital tissues are no longer produced, and organs start to fail.
All this was known, and telomeres have been a focus of anti-aging research for years. However, no silver bullets have been discovered to increase the average lifespan.
In the new study, Suh and colleagues studied Ashkenazi Jews, a homogeneous population whose genetics are well-studied. Three groups were part of the research: A very old (average age 97) but healthy group of 86 people; 175 of their offspring; and a control group of 93 offspring of parents who lived a normal lifespan.
“Our research was meant to answer two questions,” explained said Einstein researcher Gil Atzmon in a statement. “Do people who live long lives tend to have long telomeres? And if so, could variations in their genes that code for telomerase account for their long telomeres?”
“Yes” on both accounts, the scientists conclude.
The old crowd had “inherited mutant genes that make their telomerase-making system extra active and able to maintain telomere length more effectively,” the researchers write. “For the most part, these people were spared age-related diseases such as cardiovascular disease and diabetes, which cause most deaths among elderly people.”
“Our findings suggest that telomere length and variants of telomerase genes combine to help people live very long lives, perhaps by protecting them from the diseases of old age,” Suh said. “We’re now trying to understand the mechanism by which these genetic variants of telomerase maintain telomere length in centenarians. Ultimately, it may be possible to develop drugs that mimic the telomerase that our centenarians have been blessed with.”
